Tsumago-juku is the 42nd post town on the Nakasendo, located in Nagiso-machi, Kiso-gun, Nagano Prefecture. Located at the northern end of the Kiso Mountains, this post town is attractive for its tasteful cityscape that retains vestiges of the Edo period.

Tsumago-juku prospered as a transportation hub located at Oiwake of Nakasendo and Iida Kaido, and many travelers and merchants came and went. There are many traditional townhouses that have been around since the Edo period, and it has been designated as an Important Preservation District for Groups of Traditional Buildings.

The section between Magome and Tsumago on the Nakasendo is also popular as a hiking course.
